I have a 2000 frontier 6 cyl. Lately the heater will not
heat up when the the engine is running at normal operating
temp. It only blows barely warm air.

I am thinking may be the thermostat is sticking and need to
know how to replace it

Did you confirm that the top radiator hose is cool to touch? This would confirm a thermostat issue.

Replacing the thermostat:
Drain coolant
remove thermostat housing
replace thermostat, be sure to put the new one back in the same way you took the old one out, usually the spring faces downward towards the engine.
Ensure you use the thermostat gasket and apply with a sealant.
put housing back on, and fill back up with coolant.

Where exactly is the thermostat located? I see the top hose coming off the
radiator which leads back to the back of the engine, is that where it is
located?

What do you have? A 2005 Crew cab or a 2000 v6 VG engine?
I looked at my 2008, and was not able to visually see the thermostat, maybe it is behind the engine? I followed the top radiator hose and it splits in to a "Y" one hose goes to the back of the engine, and the other one goes across the front and connects to the left side of the engine block.
